The fact that formylated peptides and proteins are specific signatures of bacterial metabolism also makes them attractive targets for our innate immune system, which has adopted a strategy by which to recognize a few highly conserved non-self molecules present on or in a large group of potentially pathogenic microbes. Accordingly, the professional phagocytes (granulocytes, monocytes, macrophages) that form our first line of defense against invading microbes express such pattern recognition receptors, for which the N-formylated methionyl group is a critical determinant. Because the signals generated by the occupied formyl peptide receptors (FPR) induce chemotaxis, it has been widely held that these receptors evolved to mediate trafficking of phagocytes to sites of bacterial infection. This notion is supported by the fact that N-formyl peptides, in addition to being chemotactic, also possess other proinflammatory properties, such as the ability to activate phagocytes to release antimicrobial peptides and oxidants. The importance of proper recognition of formylated peptides is clearly illustrated by the fact that FPR deficiencies are associated with increased susceptibility to bacterial infections.
