RFLPs can be used for:
1. Identifying alleles of a particular gene, or alleles in a particular individual
2. Mapping genes on chromosomes (a gene may be linked to an RFLP marker whose location is known)
3. DNA fingerprinting
now how do we decipher this info to get the answer of the question??

because therrre is a specific gene defect ..which can be identified By RFLP.

well,this is wat my meagre knowledge about genetics say,
when u know a gene,n the point where the mutation occurs to result in a specified disease,we use PCR(or also DNA hybridisation techniques).
logic being dat,u know everything,jus u have to show dat the mutation exists,by comparing the same results with a normal or an affected population.
this we do by PCR,by jus amplifying those zones n cutting n comparing!

and in RFLP wat we do is to cut the DNA by the endonucleases,n compare if the DNA under study has more number of cuts,(or in other words wrong expression of sum gene in the sequence)
n so when we compare,these fragments,with homologus sites,we can identify the alleles(as said by pilgrim).
also,when we know the actual position of the gene,we can jus put the probe to identify the effected gene,thus mapping genes on chromosome!
the main technique being dat we compare the final FRAGMENTs n their lengths to know,if sumthing is wrong!

RFLP is generally used for those sort of diseases whose gene location is not known.or for those in which though the gene location is identified,but sum how the phenotype expression is different in different individuals.
in the above mentioned choices,all are point mutations!
only PKU is different frm the rest in being dat,it shows polymorphism(remeber 5 types of PKU exists?)
though we know dat it is on chrm 12 where the culprit is hiding,but we dunno how n wat shape or form it will take. 8)
though,RFLP can be used for the rest of the diseases also,by jus puttin a probe to identify the wrong gene,but usually,PCR is used for conditions like dat!
i wud go for PKU as answer to this question! 8)
do tell me if i went wrong anywhere!

...............RFLP is used in the diagnosis of both sickle cell disease and PKU.........the only diff. is that the gene that produces the polymorphism and the one that produces the ds. is same in sickle cell and diff in PKU.........and surprisingly RFLP is also used in the prenatal disgnosis of thalaseemia..........:
