A tough choice betwen 1 and 2...Randomization doesnt ensure that treatment and control groups are equal in all respect as such systematic loss during follow up can occur leading to selection bias...hence i will go for no. 1 as selection into trearment and control group is obviously by chance ensured by randomization..

i think ans 2 is correct.
random distribution ofbaseline characteristics and treatment between groups allows prospective assesment of intervention with out introducing bias.
one gr receive intervention under study while other one receives placebo or standard therapy,all other conditions are kept the same between groups.
