The answer is 4.

Small stretch of DNA is synthesized to be used as a probe. Northern Blot does not involve amplification of any RNA at any step. This is crucial because many times, Northern Blot is used to compare the endogenous levels of an RNA in different tissues, or differents RNAs in the same tissues.

Good luck Medicos. Some of the answers on this forum seem very naive. I pity that medical students in India do not get any research experience.

Northern blot is used to analyse dna in a given saple using the array of dna available
Southern blot is used to detect dna in a given sample ,, usually a confirmatory test
Western blot is done ot detect a specific protein in a given sample ..
