If DNA anaylsis and Karyotyping would require same thing then...
The answer to above question is Blood Monocyte....refernce Robbins 7th edition Page 170
in fetus---amniocytes and chorionic villi
in adults- Blood lymphocytes and fibroblasts

Harrisons 17th ed Chapter 64 – Methodologic Approaches to Genetic Testing
DNA testing is most commonly performed by DNA sequence analysis for mutations, although genotype can also be deduced through the study of RNA or protein (e.g., apoprotein E, hemoglobin, immunohistochemistry). The determination of DNA sequence alterations relies heavily on the use of polymerase chain reaction (PCR), which allows rapid amplification and analysis of the gene of interest. In addition, PCR enables genetic testing on minimal amounts of DNA extracted from a wide range of tissue sources including leukocytes, fibroblasts, epithelial cells in saliva or hair, and archival tissues
Robbins – Basic Pathology
Section 7 – Genetic & Paediatric Diseases – Diagnosis of Genetic Disease
Fluorescence in Situ HybridizationFISH utilizes DNA probes that recognize sequences specific to chromosomal regions. The ability of FISH to circumvent the need for dividing cells is invaluable when a rapid diagnosis is warranted (e.g., in a critically ill infant suspected of having an underlying genetic disorder). Such analysis can be performed on prenatal samples (e.g., cells obtained by amniocentesis, chorionic villus biopsy, or umbilical cord blood), peripheral blood lymphocytes, and even archival tissue sectionsAnswer: Should be Monocytes
