Normally the intracellular pH (6.9 - 7.2) is slightly less than the pH of plasma/interstitial fluid (7.4). If only this principle is taken into consideration then the basic drugs which are less ionized at higher ph would easily diffuse through membrane but since the intracellular pH is low, the drugs would ionize there and therefore get trapped. That means basic drugs should reach higher intracellular concentration......

But then wouldn't be the answer different if the site of absorption of the drug is stomach? Stomach with its low pH causes ionisation of basic drugs and they may not be absorbed intracellularly. Am I right? icon_question.gif The scenario is different after the level of duodenum.

In my reply I assumed that the drug is already in plasma because the question mentions "after absorption". Otherwise, the answer can not be predicted if the problem encompasses GI tract absorption!!
