ONE OF OUR SENIOR RESIDENT IN PATHO SAYS GLUCOSE OXIDASE IS OLD ONE.
NOW A DAYS HEXOKINASE IS USED.
I FOUND NO REFERANCE OF IT.

this is a repeat then!
It was also asked in the last december exam,where 'glucose oxidase method' was replaced with the 'GOD POD Method'(which is essentially the same).

the RxPG DNB buk also gives GOD POD as the best method.

Can any1 plz tell me the principle of Folin & wu test for Glucose. This is in regard with the Biochemistry practicals we generally do in the lab.

copper sulphate contains cupric ions which is reduced to cuprous ions in boiling conditions in presence of glucose and in fact all reducing sugars (and also non sugar reducing substances). The cuprous ions are once again oxidised to cupric ions at the expense of phosphomolybdic acid which is itself oxidised to molybdenum blue. Molybdenum blue imparts a blue colour to the medium and is estimated colorimetrically at 420 nm which gives us an estimation of the conc. of sugar.

Now heres why GOD method is the best........
1) as indicated above folin wu test will estimate not only glucose but also other reducing sugar and non sugar substances like glucoronic acid.
2)Hexokinase test not only estinates glucose but also other hexoses such as fructose and galactose
3)so by elimination glucose oxidase method id the best. the only pitfall of this method is that GOD preferentially attacks beta glucose. So if we take into account mutarotation erroneous results may be obtained if the same sample is measured after 24 hours. (This is beautifully explained in a box in vasudevan in the chapter of carbihydrate chemistry)
