Prostaglandins..and analogues.

Five majour types of prostaglandin rep..( DP, EP, FP, IP, TP)

All are G – protein coupled..

DP – Greater affinity for Pg D2
EP – Greater affinity for Pg E2 ( Enprostodil a selective agonist)
FP – For PG F2 alpha – ( Fluprostinol a sel agonist)
IP – For PG I2 ---- ( cica prostol a sel agonist)
TP – For TxA2

Thromboxin rep..

Separate for LTB4,LTC4, LTD4,

All functions through.. IP3 /DAG..

Uses .

Abortion --- PGE2.
( Dinoprost PGE2) Code foetus will die..
Induction of labour ---- PGE2,PGF2alpha(rarly)
Cervial priming – PG E2.
Post partum haemorrhage ---15 methy PGF2alpha ( Carbiprost)

Peptic ulcer…PG E1 (misoprostol, rioprostil) & PGE2 (enprostil) ( Code R.E.M sleep —Disturbed if has peptic ulcer....!)

To maintain patency of ductus. – PG E1 ( Alprstodil) ( Code--ee alpum dilation for PDA with that of in importance)

To avoid platelet damage. PGI2 (Epoprostenol) Code EPo == Ipode

Prostaglandin analogues in clinical use (MK )

ProstaglandinE1(Pg E1 analogues)

Isoprostol.

• Cytoprotective prostaglandin used in preventing peptic ulcer.
• Hass been used for cervical ripening and induction of labour.

Rioprostil.
• Cytoprotective prostaglandin used in preventing peptic ulcer.
not available in India

Alprostadil.
• To maintain ductus arteriosis patent in neonatal awaiting cardiac surgery.
• As a second line of treatment for erectile dysfunction.

ProstaglandinE2 anologue.

Dinoprostone(PGE2)
• Induction of second trimester MTP ans missed abortion.
• Termination of molar pregnancy.
• Cervical ripening.
• Induction of labour and augmentation of labour.

Enprostil(PgE2)

Cypto protective not available in India..

Prostaglantin F2 alpha.

Dinoprost.(PGF2 alpha)
• To induce 2nd TM abortion.
• Induction of labour.

Carboprost(15 Methyl PgF2 alpha)
• To induce 2nd TM MTP.
• To control post partum hemnorrhage.

Latanoprost.
• To increase uveoscleral out flow in Rx of Glaucoma
